EFFECT OF GROWTH REGULATORS COMBINATION ON MICROPROPAGATION OF FLAX LILY (Dianella tasmanica).

Abstract


Flax lily, belongs to the enormous family of Asphodelaceae. As of their fascinating and charming properties, lilies are in a huge market demand in Pakistan. The aim of this research was to design an efficient combination of growth regulators in MS medium for the rapid growth of Dianella tasmanica (Flax Lily). In this study different combinations of growth regulators for shooting and rooting of lily were investigated. The multiplication was achieved and the highest length of shoots (1.8 cm) was observed on M4 medium containing MS medium fortified with BAP (2.0 mg/l), IBA (1.0 mg/l), IAA (1.0 mg/l) along with sucrose 40gm/l and 10% coconut water while the significant length (3.5 cm) was obtained on M2 medium containing MS medium supplemented with BAP (2.0 mg/l), IBA (1.0 mg/l), IAA (1.0 mg/l) supplemented with sucrose 0gm/l along with 10% coconut water and also on M5 medium containing sucrose 60gm/l in MS basal medium supplemented with BAP (2.0 mg/l), IBA (1.0 mg/l), IAA (1.0 mg/l) added with 10% coconut water. For rooting of Flax lily shoots, MS4 medium containing 3mg/l Indole-3-Butyric Acid (IBA) along with constant 6-Benzylaminopurine (BAP) 2mg/l and Indole-3-Acetic Acid (IAA) 1mg/l supplemented with 0.5gm/l activated charcoal was used and resulted in the highest number of roots (6).The significant length of roots (2.5 cm) was obtained on MS6 medium containing 5mg/l IBA along with constant (BAP) 2mg/l and (IAA) 1mg/l supplemented with 0.5gm/l activated charcoal. In this study multiple bulbs were protruded from single initiated bulb and formed many shoots on multiplication medium, rooting was also successfully achieved, and plants were transferred into soil after two months of rooting. Acclimatization was initially performed in a soil under controlled temperature at 25±2 ºC with 16/8 hrs photoperiod. After 30 days plantlets were transferred into pots and were placed in a green house. In this study Flax lily (Dianella tasmanica) was successfully micropropagated through plant tissue culture technology.
