Effects of Piriformospora indica inoculation on growth and changes in transcriptome in Brassica chinensis seedlings supplied with selenate

Abstract
Selenium (Se) is an important trace element for human health and is related to many human diseases. Selenium biofortification is a good way to satisfy the demand of Se daily uptake for human health. In this present study, seedlings of Chinese cabbage (Brassica chinensis L.) were inoculated with root endophytic fungus Piriformospora indica (syn: Serendipita indica, here abbreviated as Piin) under selenate application in soil, and four treaments occurred (i.e., CK, Se, Piin, Piin+Se treatment). Growth and quality and the Se-related transcriptome was analyzed. The results showed Piin+Se treatment did not significantly influence total chlorophyll and carotenoids, but significantly increased net photosynthesis rate, compared with control (i.e., CK treatment), resulting in a significant increase in shoot and root biomass. Under Piin+Se treatment, Chinese cabbage seedlings contained significantly higher levels of anthocyanin, ascorbate, soluble proteins and sugars, phenols, flavonoids, compared with control. However, the treatment did not affect glutathione levels. Piin+Se treatment resulted in a significant increase in organic and inorganic Se accumulation, compared with control. Transcriptome analysis showed that Piin+Se treatment affected the expression of genes, especially those related to secondary metabolism and Se metabolism. These changes in secondary metabolism and Se metabolism were closely related to phenols, flavonoids, anthocyanin, ascorbate, and Se accumulation. Thus, it could be concluded that P. indica inoculation in combination with selenate fertilizer improved quality and Se accumulation in Chinese cabbage seedlings. Given the high consumption of Chinese cabbage seedlings in China, the combination of P. indica inoculation and selenate fertilizer should be used in cultivation of Chinese cabbage to satisfy the demand for Se daily uptake because of the long-term effect of the fungus in field.
