Abstract
This study was conducted to check the antifungal and Ochratoxin A (OTA) degradation potential of soil Borne Bacillus species. Ochratoxin A (OTA) is one of the most common mycotoxins. It is found in variety of food/feed and food/feed commodities ranging from cereals to spices. Different physical, mechanical, physicochemical and chemical methods are being employed to inactivate and reduce the content of mycotoxins in feed/food. However, these approaches did not prove as efficient and effective due to reduce nutritive value, palatability and half-life of food. Bacillus species were isolated from the soil samples which were collected from different agricultural and farms soils in and around Lahore. These 21 isolates out of 40 were processed for antifungal and Ochratoxin A degradation potential. Antifungal testing of these isolates was performed by well diffusion assay and no zone of inhibition and hence no antifungal activity was observed. The cells of Bacillus was divided into Cell free supernatant (CFS), Intracellular Fraction (IF) and Cell pellet (CP) for checking OTA degradation potential of these fractions. The binding and degradation of OTA with these fractions were checked and analyzed by Thin Layer Chromatography (TLC) which showed significant binding and degradation of OTA. The isolates whose fractions showed significant binding of OTA were processed for DNA extraction, Polymerase Chain reaction (PCR) and sent for sequencing. Sequenced Bacillus isolates were Bacillus rugosus, Bacillus cabrialesii, Bacillus teqilensis, Bacillus subtilis, Bacillus niacin and Bacillus betaviensis.
