Effect on the Root of Wheat Seedlings Exposed to Enhanced UV-B Radiation

Abstract
Enhanced UV-B radiation have various effects on wheat growth morphological, physiological, biochemical and genetic mechanism. The roots of wheat seedlings treated different days with UV-B was used as experimental material. This experiment analysed UV-B effect on the number of adventitious roots, average root length, root biomass, root vigor of wheat, and the UV-B radiation had different degrees of influence on wheat roots. Then, screening the best way to extract root protein from Phenyl-methanol/ammonium acetate precipitate, TCA/acetone method, Urea thiourea method, while TCA/acetone precipitation was the best way to extract root protein by SDS-PAGE. Finally, the root of wheat dealing with UV-B was used to extract protein in TCA/acetone precipitation, and it is concluded that protein content in ultraviolet-B treatment group (B) was higher than that of control group (CK) at the early stage of the radiation, with the increase of irradiation time the protein content of B was lower than that of CK.
