EFFECT OF DIFFERENT MUTAGENSIS ON MICROSTRUTURE AND ULTRASTRUTURE OF ALFALFA

Abstract
The objective of this study is to research the effect of Gongnong 1, Wega7F, WL319HQ, Aohan were treated by 60COγ-ray irradiation, ultraviolet, Ethyl methyl sulfonate(EMS) and magnetic field-free space on microstructure and ultrastructure of leaf. Screening the optimal induction mutation method and breeding new alfalfa varieties for alpine region. Gongnong 1, Wega7F, WL319HQ, Aohan as the test materials. 60COγ-ray irradiation(150, 300, 450 Gy), ultraviolet(30, 60, 90 min), EMS(0.1%, 0.2%, 0.4% (v/v)) and magnetic field-free space(180 d) were set. Microstructure and ultrastructure of leaf were determined. The microstructure of the leaf thickness was increased, which Gongnong 1 under 0.4% (v/v) was the highest, as much as 446.16μm with an increase of 25.86. Meanwhile, the leaf vein protuberant degree was decreased by the different mutagenic. Cell tight degree of Gongnong 1 induced by 60COγ-ray irradiation and ultraviolet more than the CK. wega7F cell become more closer under all mutagenic treatments. The ultramicrostructure of the chloroplast in the alfalfa leaves was observed under different mutagenic. Lower dose or concentration of mutagenic treatments, the chloroplast of the cell showed distortion, ambiguity in membrane,loose in grana lamella,disintegration in thylakoid, increases in lipid ball. Under the highest dose or concentration mutagenic treatment and magnetic field- free space, the chloroplast of the cell displayed swelling and distortion, fracture in grana lamella and disintegration in membrane system. Membrane systems of the chloroplast were disintegrated and its chloroplast structures were damaged more seriously, both plastids and starch grain were increased. Mutagenic efficiency of 60COγ-ray irradiation and EMS were higher, mutants materials were more abundant, simple operation, which were the reasonable mutation methods. 150Gy 60COγ-ray irradiation, 60min ultraviolet and 0.4% EMS were the better mutagenic treatments that were conducive to screening mutants.
