Genotoxicity of silver nanoparticle in wheat root tip cells

Abstract


Nowadays, the distribution and use of nanoparticles increased quickly through the current years, while the knowledge about mode of action, ecological tolerance, and biodegradability of these chemicals is still insufficient. Wheat (Triticum aestivum L.). is the most important crop in worldwide. Two different sources, green and chemical synthesis of nano silver (AgNPs) was investigated in the current research to study the effect of these materials on chromosomal aberrations and cell division. The results demonstrated that Nano silver might arrive easily into the cells and interfere with cell’s normal function. Three different expose time and four AgNPs concentrations were used. four AgNPs 10, 20, 40 and 50 ppm and wheat root tips were treated after 8, 16 and 24 hrs. The different concentrations of nanoparticles were added to the tested grains until the root length retched from 1.5 to 2 cm in length. Mitotic index (%) was measured from ~ 2000 cells for each concentration. The treated root-tip cells exhibited various types of chromosomal aberrations as orientation at metaphase, breaking of chromosomes, metaphasic plate distortion, spindle dysfunctioning, stickiness, precocious movement at metaphase and bridge, fragmentation scattering, unequal separation, multiple bridge, fragmentation, scattering, laggard and elongation, gap chromosome, multi polar anaphase, erosion, distributed chromosome and lagging chromosome during the current research.
