SEQUENCING AND IN SILICO ANALYSIS OF OsGLP12-3 PROMOTER REGION IN DIFFERENT VARITIES OF ORYZA SATIVA

Abstract
Germin and germin-like proteins (GLPs) is a class of plant proteins that belongs to super family “Cupins”. They are reported to express in different plants organs like nectar glands, roots, leaves and seeds. Gene expression of germins and germin like proteins is also been reported to be included in plant defense against pathogenic attack and in response to different environmental stress conditions. In this study, promoter analysis of OsGLP12-3 promoter region is carried out in three rice varieties as it explains functioning, regulation and response of the particular gene. The OsGLP12-3 promoter region, 2kb, was amplified using genomic DNA of three varieties of Oryza sativa i.e. Nipponbare, Swat-1 and Basmati-2008. The amplified products were purified and quantified and then subjected for sequencing. In Silico comparative analysis of the sequenced results of all the three varieties was done using online tools PlantCARE database. Different 39 cis-acting regulatory elements were identified in the OsGLP12-3 promoter region having diverse range of functions. Each element was found to exist with distant copy number and distant location in all the three varieties of rice. In the study, OsGLP12-3 promoter region was found to contain enormous upstream regulatory elements confined to give defense against biotic and abiotic stresses. Upstream regulatory region (promoter) of OsGLP12-3 was assessed using different bioinformatics tools to find out regulatory aspect of the particular promoter
