Analysis on phenotype, catalpol accumulation and methylation of Rehmannia glutinosa

Abstract
Rehmannia glutinosa is one of the traditional Chinese herbal medicines, phenotype, catalpol accumulation, genomic methylation of Rehmannia glutinosa planted in Wenxian and Xinxiang were studied in this research. During the growth and development of Rehmannia glutinosa, leaf length, plant height and catalpol accumulation of Rehmannia glutinosa increased continuously along with the extension of growth stage, and reached the peak in the later growth stage of Rehmannia glutinosa. In addition, 5mC content and methylation level in genome also increased along with the growth of Rehmannia glutinosa, and reached the highest value in the later growth stage. As compared with that of Rehmannia glutinosa planted in Xinxiang, leaf length, plant height, catalpol accumulation and 5mC content of Rehmannia glutinosa planted in Wenxian were all higher at the same growth stage of Rehmannia glutinosa, the genomic methylation level was also higher at the later growth stage of Rehmannia glutinosa, yet was lower at the early growth stage.Therefore, these research results would help to explore the correlation among ecological environment, DNA methylation and growth of Rehmannia glutinosa, and Wenxian maybe more suitable for planting of Rehmannia glutinosa.
