CHARACTERIZATION OF LACCASE PRODUCED BY A NOVEL STRAIN OF FOMES FOMENTARIUS IN SOLID STATE FERMENTATION
Abstract
A local isolate of Fomes fomentarius produced high titters (260 U/mL) of laccase after incubation at 35C for 8 days on corn stover solid medium. Optimal laccase activity (240 U/mL) was obtained at pH 7 and it was increased to (258-260 U/mL) at 35-40C temperature. Laccase was fractionated by 50% saturation with (NH4)2SO4 having 1.5 purification fold after dialysis from membrane. Gel filtration increased the purification factor up to 2.2 fold with 1.65 U/mg specific activity. The laccase had an estimated molecular weight of 44kDa when characterized with Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis process. Laccase revealed maximum activity (230 U/mL) for 2,2 azino bis 3 ethylthiazoline 6 sulphate (ABTS) and oxidization of ABTS showed kinetic constants Km of 1 mM with Vmax values of 60 mM/minute. Purified laccase at a concentration of 0.25 – 1.00mL/mL of lignin led to 80 – 90% lignin degradation. Laccase produced through Fomes fomentarius revealed good decolorization property (82%). Thus, Fomes fomentarius laccases can potentially be employed in paper, textile, pulp, food industries and also used in waste detoxification.
