Regulatory inflammatory response of Safflower injection in treating Stroke by ERS

Abstract
Safflower injection has the effective treatment of resisting ischemia reperfusion injury, which is used in the treatment of ischemic stroke. This experiment was conducted to explore the effect of safflower injection in the treatment of stroke modulating inflammation through ERS by MCAO model rats, provide some experimental basis for the clinical application of safflower injection in the prevention and treatment of cerebral infarction. We established MCAO model rats by modified suture method, injected intravenously safflower injection. Immunohistochemistry was used to detect the expression of ERS markers CHOP and GRP78. The expression of GRP78 mRNA and CHOP mRNA was detected and the level of high-sensitivity C-reactive protein (CRP) by ELISA. In the research, the area of cerebral infarction in the safflower injection group was significantly smaller than that in the model group, the score of nerve injury was significantly lower in the safflower group than the model group, the expression of GRP78 positive cells in the safflower injection group was significantly higher than that in the model group, while the expression of CHOP in the safflower injection group was significantly lower than that in the model group. The CRP level of the safflower injection group was significantly lower than that of the model group. The study display the Safflower injection has protective effect on stroke involved ERS related to regulation of GRP78 and down regulation of CHOP. The CRP also may play a role in the prevention and treatment of ischemic stroke.
