Production of Vicia faba Interspecific Hybrid Through In Vitro Propagation Technique
Diaa Ayoub Ibrahim
Abstract
Abstract Embryo rescue of the hybrid derived from two Vicia faba lines (ILB 1270, Giza blanca ILB 1814) was studded under in vitro conditions. Four protocols (A,B,C and D) were used for inducing embryonic structures. The results indicated that the highest effective response was recorded by using (B) protocol which recorded the highest number of new embryonic structures. Meanwhile, the results clarified that adding Kinetin to MS medium with B5 Vitamins (MS basal salts supplemented with B5 Vitamins + 2.3 mgl-1 Kinetin + 20 gml-1 sucrose and 8 gml-1 Agar) encourage shoot proliferation and elongation. in vitro flowering was achieved by producing large numbers of flowers when cultured in DKW medium supplemented with 1 gml-1 MES, 30 gml-1 sucrose and 2 gml-1 gelrite. On the other hand, the results revealed that 5% of regenerated shoots were able to form roots by using half strength of MS medium salts + 2.5 mgl-1 ascorbic acid + 1.0 mgl-1 PVP + 15 gm-1 sucrose + 2 gml-1 phytagel.
