Antipyretic activity of Various Fractions from Nigella sativa Methanolic Extracts and its Mass Spectrometry Analysis

Nigella sativa, an annual flowering plant of family Ranunculaceae, is native to Mediterranean region. The reproductive plant parts (seeds/oil) have been extensively utilized for pharmaceutical purposes. Therefore, it was planned to determine the strength of both vegetative and reproductive parts to get a natural potential pharmaceutical, for pyrexia. This plant is well known for food and medicinal uses. The use of its seeds and oil is common for treatment of many diseases, including arthritis, hepatitis, asthma, diabetes, inflammatory diseases, cardiac disorders and digestive disorders. N.sativa vegetative/reproductive parts were extracted in methanol, chloroform, n-hexane, ethyl acetate, n-butanol and distilled water. The stock solutions were double diluted like 200mg/ml, 100, 50, 25, 12.5, 6.25, 3.125 and 1.625mg/ml in DMSO. The mice weighing 25-30 gm were used for activity and were pre-domesticated for 5 days in the animal house of university of veterinary and animal sciences (UVAS) Lahore. Pyrexia was induced by subcutaneous injection of brewer’s yeast (10ml/kg) in distilled water. The control group contained mice treated with standard medicine paracetamol whereas in the remaining six groups mice were treated with plant extract concentrations. Results showed that N.sativa had a concentration dependent antipyretic effect on mice. The rectal temperature decreased significantly with the increasing concentration of the extracts for 0.5-4hrs of oral administration. The methanolic fraction of reproductive part showed marked reduction in rectal temperature that was more significant than standard paracetamol at 200 mg/ml followed by n-hexane, n-butanol and ethyl acetate extracts. GC-MS analysis of the potential extracts was also performed.
