ESTIMATION OF GENETIC DIVERSITY IN DIALLEL CROSSES OF MAIZE GENOTYPES USING SIMPLE SEQUENCE REPEAT (SSR) PRIMER SETS

Abstract


Genetic diversity is an important tool for vigor maize breeding. SSR markers were indicated to be a best element for tracing of genetic diversity in maize genotypes. The experimental maize project was patronize to estimate genetic diversity among 36 maize genotypes using 3 simple sequence repeat (SSR) primer-sets. The array of genetic diversity was <100-850> bp. Total number of 297 bands was shown in PCR products. Out of 297 bands 104 bands were shown in primer (Um-2281), 103 bands were shown in primera (Umc 1297) and 90 bands were shown in primer (Umc 1256). Out of these three primers only (Umc1256) exhibit only one monomorphic band and the remaining all exhibit polymorphic bands. All bands of SSR primers ranged from 100bp to 850 bp. Cluster analysis was conducted to assess the genetic similarity (GS) among genotypes while principal coordinate analysis was conducted to estimate the genetic variation and conformation of results. Cluster-1 contains 10 genotypes which again sub divided into two sub-clusters (A) and (B). Cluster-1 indicated high level of genetic diversity as compared the remaining clusters. Cluster analysis exhibited clear variation and diversity in the maize genotypes. The maize genotypes which exhibit maximum variation showed maximum diversity that are recommended for coming maize breeding programs.
