GENETIC FIDELITY ANALYSIS OF MICROPROPAGATED PERIWINKLE (CATHARANTHUS ROSEUS L.) WITH INTER SIMPLE SEQUENCE REPEAT (ISSR) MARKERS

Abstract


Catharanthus roseus is important medicinal plants as the entire plant contain high amounts of alkaloids. Shoot tips and internode segments were used in Murashige and Skoog (MS) medium supplemented with different concentrations of 6-benzylaminopurine (BAP), Gibberellic acid (GA3), and Indol-3-butyric acid (IBA) hormones. The Highest establishment rates (80%) obtained in the MS medium containing 1.0 mg/L GA3 and BAP. The Highest proliferation rate (5.20 shoots/explant) and average shoots length (6.30 cm) also obtained in 1.5 mg/L BAP with 0.5 mg/L IBA, respectively. In addition, the best rooting response (85%) was observed on 1/4 MS containing 1 mg/L IBA. Regenerated plantlets were successfully acclimatized and hardened in a culture room and then transferred in a greenhouse; the plantlets showed a 90% survival rate. Subsequently, the regenerated plants genetic homogeny we verified with different Inter Simple Sequence Repeat (ISSR) markers. Results of our study added new knowledge to growing body of Madagascar periwinkle large scale propagation by in vitro techniques.
