CLONING AND EXPRESSION ANALYSIS OF A NOVEL ZINC TRANSPORTER GENE FROM NASTURTIUM¡¡OFFICINALE

Abstract


ZIP (Zinc-regulated transporter, Iron-regulated transporter like Protein, ZIP) is a kind of zinc and iron transporter family. It plays an important role in uptaking or transporting zinc or iron in plants. Herein, a novel zinc transporter gene NoZIP3 was cloned from Nasturtium officinale root using rapid-amplification of cDNA ends (RACE) method and the sequence analyzed by bioinformatics. The function of NoZIP3 was assessed by functional complementation assay and the expression determined by quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR). The full-length cDNA was 1029 bp, encoding a polypeptide of 342 amino acids with a conserved domain of the typical ZIPs. The sequence of NoZIP3 shared a high similarity (57¨C89%) with that of the other plants. NoZIP3 could complement the mutant yeast deficiency in zinc transportation; however, it could not complement that of iron transportation. qRT-PCR revealed that the expression of NoZIP3 was significantly induced both in roots and leaves by zinc or iron deficiency. These results provided the theoretical foundation for rational consumption and scientific cultivation of this plant.
