THE OVEREXPRESSON OF CO-ACCC FROM CAMELLIA OLEIFERA IN ARABIDOPSIS THALIANA
Baoming Wang
Abstract
In this study, the pCAMBIA1304-35S was used to conduct over-expression vector of Co-accC from Camellia oleifera through the media pMD18-T vector. The recombinant was confirmed by PCR and double digestion. It was transformed to Arabidopsis plants through pollen tube pathway mediated by Agrobacterium tumefaciens GV3101, and transformed seeds were screened by PCR and double digestion, and the T0 and T1 generation Arabidopsis plants were identified by Basta. 24 plants and 17 plants transformed plants were finally obtained, respectively. Our study has explored the Co- pCAMBIA1304-35S-accC overexpression through agrobacterium tumefaciens, and confirmed that it was an efficient tool for gene overexpression in plants, and will be beneficial for revealing gene’s function and increasing camellia oil yields of through genetic engineering.
