WESTERN BLOT METHOD WITH P6 TO DETECT SOUNTHERN RICE BLACK-STREAKED DWARF VIRUS

Abstract


Southern rice black streaked dwarf virus (SRBSDV) is a severe threat to both the yield and quality of rice products worldwide. Traditional detection methods for the diagnosis of SRBSDV are convenient. However, these methods increase the number of false positives. Western blot can precisely solve the problem. In this study, P6, a viral RNA-silencing suppressor, was expressed and purified in vitro. Two polyclonal P6 antibodies were obtained and valued by ELISA and WB analysis. Then, a WB assay was developed for the identification of SRBSDV antigens derived from suspected rice samples collected from 9 districts. The assay results showed that Libo, Pingtang, Huishui, Dushan, and Anshun had an SRBSDV outbreak. Moreover, the virus content was quantified in rice tissues based on WB analysis. Our results revealed that SRBSDV mainly accumulated more in rice stems than leaves. The SRBSDV P6 antibody can be used for SRBSDV detection and monitoring.
