PREVALNCE, IDENTIFICATION, AND CHARACTERIZATION OF MACROFUNGI BELONGING TO GENERA TREMETES AND AGROCYBE (AGARICOMYCETES) IN MANSEHRA REGION KPK PAKISTAN.

Abstract
The morphological and molecular characterization for identification of 18 species belonging to two genera of macro fungi (Basidiomycota) was done for the purpose of hunting the surrounding areas including Oghi, Balakot, Ichrian, Kotkay,Bafa,Jabori, Sumb, khaki and Attarshesha were visited and explored for collection. The morpholgical data was taken on the collected site and properly snapped with the substrate and safely observed for characteristics in microbiology research laboratory, Hazara University Mansehra. DNA of the collected species were extracted by kit method and ITS4 primer designed from conserved regions described in previous studies was used for PCR. The amplified PCR products were sequenced from Macrogen Korea via worldwide scientific Pakistan. Phylogenetic analysis of the obtained sequences was done on the basis of maximum likelihood method using Mega 6.0. Morphological examination showed that two genera were found frequently in different areas including Tremetes and Aqrocybe. Different species of the identified genera included i-e T. gibbosa, T. hirsute, T. cubensis,T. manilaensis,T. vesicolor,T. elegans, A. aegerita, A. sp, A. pediales, A. cylindra. All the species were submitted to gene bank and obtained accessation number. Our findings based on phylogenetic analysis showed the existence of few new species in the already described genera as well as some novel species from the region. Based on our results, we concluded that the mountainous area, grass land including watery area or damp places of district Mansehra are enriched with Polyporales and Agaricales species and can be collected in different period of time throughout the year. For more exploration and diversification research is open.
