Study of Dreb2 gene expression and Drought stress tolerance on Sensitive and Resistant durum wheat genotypes (Triticum durum L.) under drought stress

Abstract
To consider the resistance of renitent and sensitive genotypes of durum wheat under drought stress an experiment was carried out in cold region of Ardabil on 6 genotypes of durum wheat with Iran and Azarbayejan sources. At first, experiments were done separately under water and drought stresses in completely accidental plots. The results of variance analysis showed that there are significant differences between genotypes yield’s at 1% of probable level. This indicates that high genetic varieties are crucial to choose the best genotype regarding its characteristics. Using drought stress indexes of genotypes, they can be sieved according to their drought stress. On the basis of STI index that is considered as the most valid index for stress resistance, Leucurum (Langan), Leucurum (Germi) and Apulicum (11010) are the most resistant genotypes respectively. Among these genotypes, Apulicum (11010) var. responds to environmental stress more than other varieties, has favorable sustainability of operation and has higher mean of components operation. The results of germination percentage showed that Leucurum (Langan), Leucurum (Germi) and Apulicum (11010) had the best germination level comparing with other varieties. The results of Real Time PCR indicated that there is a significant difference in Dreb2 gene in terms of considered genotypes at 1% level. The results of mean comparison in Duncan method indicated that in Dreb2 gene, Genotype No. 3 that is Melanopus (Goliblag) was the best variety at 5% level, because it and the most expression and got the A rank. The results of correlation showed that there is a significant and negative relationship between Dreb2 gene expression and STi index. Regarding the results, it seems that the gene expression of Dreb2 couldn't have been the stress resistance index of genotypes. In terms of gene expression, sensitive varieties had the most level of index. Perhaps, varieties have tried to resist against stresses but the genes couldn't play proper role and the sensitivity of other genes forego comparing with this gene and this gene wasn't probably effective at this step, but it can be effective in the other stages.
