ASSESMENT OF GENETIC DIVERSITY AMONG DIFFERENT GENOTYPES OF Solanum lycopersicum L. USING RAPDs AND SSR MARKERS

Abstract
The study of genetic diversity in landrace collections is important for germplasms conservation. In addition, the characterization of much diversified materials with molecular markers offers a unique opportunity to define significant biological and agronomic interest. This study was carried out on eleven different lines of Solanum lycopersicum L (tomato) specie in order to assess the diversity in their genomes using RAPD (random amplified polymorphic DNA) and SSR (simple sequence repeats). The procedure involved DNA extraction by CTAB method, PCR and finally gel electrophoresis. The maximum number of bands amplified among all the RAPD primers was B-06 which gave a total of 32 bands and showed 100% polymorphism. Minimum number of bands obtained was by H-09 which gave a total of 9 bands and showed 18% polymorphism. Cluster analysis showed maximum similarity in 19858, 19903, 19902, 19894, 19889, 19887 and minimum similarity in 34168. Percent polymorphism was 75%. In case of SSRs total numbers of bands obtained were 51. TSSRE5 gave 8 bands in 11 lines and TSSRB2 gave 10 bands out of 11 lines. However TSSRA1, TSSRC3 and TSSRD4 gave bands in all the eleven lines. Genotypes which showed maximum number of bands were 19899, 17883, 29448, 34168, 19902, 19858 and 17882. The dendrogram constructed showed the maximum similarity obtained by SSR markers was in 19858, 19903, 19902, 19894, 19889, 19887 and minimum similarity observed was in 34168. The use of molecular markers is useful in identifying genetic diversity in tomatoes and this information can be used in future breeding programmers.
