In tissue culture regeneration potencial of Malacocarpus crithmifolius from Mangyshlak (Kazakhstan)

Abstract


Malacocarpus crithmifolius is an endangered Iranian-South Turonian relict species with a low population size and a limited range of sprouting. The purpose of this study was to initiate in vitro and micropropagation of Malacocarpus crithmifolius. The explants of vegetative and generative shoots and Murashige & Skoog medium (MS) with 6-benzylaminopurine (BAP), indole-3-acetic acid (IAA), naphthyl acetic acid (NAA), 2,4-dichlorophenoxyacetic acid (2,4-D), and indole-butyric acid (IBA) were used. The type of organogenesis in regard to explants and the effects of hormonal treatments on shoot formation frequency, the average number of shoots, callusogenesis, and shoot rooting were investigated. Direct organogenesis was observed only at generative explants on the MS medium contained BAP in combination with NAA or IAA. The bud conglomerate was initiated in leaf axil under inflorescences after week cultivation on induced medium. Multiple shoot formation was reached by multiple sub-cultivation of revealed buds and adventitious shoots. The multiplication rate at MS with 4 mg/L BAP + 0.25 mg/L NAA and at 1.5 mg/L BAP + 0.1 mg/L IAA composed in mean 5.85 for three passages after 12-weeks culture. In conclusion, the optimal explant and hormonal medium for initiation and micropropagation of M. crithmifolius were developed.
